The Expressions of NFBD1 in Early Cervical Squamous Cell Carcinoma and CIN

Objective: Detection NFBD1 (nuclear factor with BRCTdomains protein 1) in normal cervical tissue, CIN (including carcinoma in situ) and cervical squamous cell carcinoma (I \u0026 II period) in the expression and significance, preliminary study NFBD1 in normal cervical tissue development precancerous lesions to the relevance of the process of the development of cervical squamous cell carcinoma. Methods: Clinical and pathological data of patients with complete cervical tissue, 90 cases, including 30 cases of normal cervical tissue, CIN organizations (including carcinoma in situ) of 30 cases of cervical squamous cell carcinoma (I \u0026 II period) 30 cases (all cases in radiation therapy and drug chemotherapy) were not drawn before. Immunohistochemical SP method and RT-PCR were used to detect NFBD1 protein and NFBD1 mRNA expression in each group. Results: 1. Immunohistochemistry results show: NFBD1 in normal cervical tissue, CIN group (including carcinoma in situ) squamous cell carcinoma of the cervix were expressed, the immunohistochemical positive products are mainly located in the nucleus, the small number of cytoplasmic coloring . With cervical lesions progress, NFBD1 protein staining positive cell rate gradually increased staining intensity gradually increased, immunohistochemistry score gradually increased. (1) CIN the group NFBD1 protein immunohistochemistry score: CIN Ⅰ higher than normal cervical tissue, below CINII, P values ??were 0.50,0.08, there was no statistically significant (P = gt; 0.05). Higher than CIN II, CIN Ⅲ P value of 0.04 was statistically significant (P lt; 0.05). Higher than normal cervical tissue, CIN Ⅱ P value of 0.00 was statistically significant (P lt; 0.05). Cervical squamous cell carcinoma NFBD1 protein immunohistochemistry score: With the staging of increased, NFBD1 expression gradually increased cervical squamous cell carcinoma Phase Ⅱ gt; cervical squamous cell carcinoma of I gt; CIN Ⅲ P are 0.04, statistically significant (P lt; 0.05). Cervical cancer the Ⅰ higher than normal cervical tissue, P 0.00, statistically significant (P lt; 0.05). (2) in cervical squamous cell carcinoma group, NFBD1 protein expression and pathological grade. With the pathological grade higher cervical squamous cell carcinoma, NFBD1 enhanced expression of cervical squamous cell carcinoma the grade Ⅲ gt; the cervical squamous grade Ⅱ gt; Ⅰ grade cervical squamous cell carcinoma, are 0.04, statistically significant (P lt; 0.05) . 2. NFBD1 mRNA RT-PCR test results show that: NFBD1 mRNA expression and cervical squamous cell carcinoma (I \u0026 II period) in normal cervical tissue, CIN (carcinoma in situ), basic conclusions with NFBD1 immunohistochemistry conclusions consistent. NFBD1 mRNA in normal cervical tissue, CIN and cervical squamous cell carcinoma (I \u0026 II period), with cervical lesions progress, NFBD1 expression gradually increased. RT-PCR results showed that: (1) CIN group higher than normal cervical tissue, CIN Ⅰ below CIN Ⅱ, P values ??were 0.21,0.83, there was no significant statistical significance (P> 0.05). CIN Ⅲ was higher than CIN Ⅱ, p value of 0.00 was statistically significant (P lt; 0.05). Higher than normal cervical tissue, CIN Ⅱ P value of 0.00 was statistically significant (P <0.05). Cervical squamous cell carcinoma NFBD1 mRNA RT-PCR test results show that: the installments increase, NFBD1 mRNA relative expression gradually increased cervical squamous cell carcinoma II period gt; cervical squamous cell carcinoma of I gt; CIN Ⅲ P values ??were 0.03, 0.04, were statistically significant (P lt; 0.05). Cervical squamous cell carcinoma stage Ⅰ higher than normal cervical tissue, P 0.00 was statistically significant (P lt; 0.05). (2) in cervical squamous cell carcinoma, the relative expression of mRNA NFBD1 with pathological grade. With cervical squamous cell carcinoma pathological grade increased, the relative expression of mRNA NFBD1 enhanced. Cervical squamous cell carcinoma the grade Ⅲ gt; cervical squamous cell grade Ⅱ gt; the grade Ⅰ of cervical squamous cell carcinoma, P were 0.03 and 0.02, statistically significant (P lt; 0.05). Conclusion: Immunohistochemical detection of conclusions and RT-PCR detection conclusion is basically the same, normal cervical tissue, CIN (including carcinoma in situ) and cervical squamous cell carcinoma were expressed NFBD1 day with cervical lesions progress, NFBD1 expression also elevated; cervical squamous cell carcinoma, With the the the cervical squamous pathological grade increased NFBD1 of expression is also elevated. The speculated NFBD1 expression abnormalities may be associated with cervical cancer and development.
