Effect of Ursolic Acid on Apoptosis and the Expression of Survivin Protei on Bladder Cancer 5637 Cell Line

ObjectiveTo observe the effects of ursolic acid(UA) on high-risk superficial bladder cancer cells (5637 cell line) in vitro,Including the cell proliferation and induce the apoptosis of interaction, For bladder cancer treatment for Ursolic acid provides the basis.Methods5637 cells cultured in vitro conventional, and logarithmic growth phase cells to divided into two groups randomly. The experimental group joined the mass concentration10, 20, 40, 80ug/ml of UA were treated 24h,48h; the control group without ursolic acid,and the establishment of negative control group. Using the MTT method detection each cell proliferation inhibition rate; inverted phase contrast microscope observe the apoptotic cells, Flow cytometry was used to detect the apoptosis rate. Western - bolt method to detect Survivin protein expression.ResultsDetermined by MTT results show that 10 ~ 80 ug/ml Ursolic acid could restrain the 5637 cell proliferation effectively, inhibition is time-dose dependent, 80 ug/ml role for 48h group’s inhibition rate is 47.64 + 1.39% (P < 0.05); Nverted phase contrast microscope visible cell volume decreases, nuclear become pyknotic, cytoplasmic appear empty bubble, coated shrinks and apoptotic body formation; Flow cytometric analysis cells tests detect, the experimental cells present apoptosis, and apoptosis rate as drug concentration and duration increase. 80 ug/ml role for 48h group’s apoptosis rate is 45.20±1.12% (P < 0.05); Western-blot results show that UA could reduce Survivin protein expression level, and as the drug concentration and duration increased decline(P < 0.05).ConclusionsUrsolic acid for bladder cancer 5637 cells have obviously the role of inhibiting proliferation, and also have apoptosis-oriented role. Its mechanism may be reduce Survivin protein expression and apoptosis to fulfill. UA may become the new clinical medication treatment of bladder cancer.
