Effects of Hyaluronic Acid on Osteopontin and CD44 of Fibroblast-like Synoviocytes in Patients with Osteoarthritis of the Knee

Objective:This study is designed to change the contents of HA in the fibroblast-like synoviocytes, in order to investigate the effect of HA on the expression of OPN and CD44 of fibroblast-like synoviocytes in patients with osteoarthritis of the knee.Methods:The study was performed on 10 specimens of synovium or synovial fluid obtained from osteoarthritic patients who underment total knee arthoplasty or arthroscopic surgery. Then all the synovial samples were cultured and purified to obtain fibroblast-like synoviocytes in vitro,take the 4-6 passaged cells into three groups:(1)the control group,(2)treated with 100 mg/ml HA for 24 h, (3)treated with 300 mg/ml HYL for 24 h, We designed and validated the real-time quantitative polymerase chain reaction (Q-PCR) assays with SYBR Green dyes for simultaneous quantification of the expression of the OPN mRNA and CD44 mRNA. All results statistics were analysed by SPSS 17.0.Results:1 Compared to controls, HA (100ug/ml) up-regulated OPN mRNA expression in OA fibroblast-like synoviocytes, and the difference showed statistically significant.(p<0.05).2 Compared to controls, HYL (300ug/ml) down-regulated OPN mRNA expression in OA fibroblast-like synoviocytes, and the difference showed statistically significant.(p<0.05).3 The difference of OPN mRNA expression between HYL (300ug/ml) and HA (100ug/ml) interferenced in OA fibroblast-like synoviocytes,showed statistically significant. (p<0.05). 4 Compared to controls, after HA (100ug/ml) interferenced, the CD44 mRNA expression in OA fibroblast-like synoviocytes, showed no difference of statistically significant.(p>0.05).5 Compared to controls, after HYL(300ug/ml) interferenced, the CD44 mRNA expression in OA fibroblast-like synoviocytes, showed no difference of statistically significant.(p>0.05).6 The difference of CD44 mRNA expression between HYL (300ug/ml) and HA (100ug/ml) interferenced in OA fibroblast-like synoviocytes,showed statistically significant. (p>0.05).Conclusion:1 HA can up-regulate OPN mRNA expression in OA fibroblast-like synoviocytes,the high expression of OPN mRNA in OA fibroblast-like synoviocytes may be a result of inceased HA level of OA synovitis.2 HA can not affect CD44 mRNA expression in OA fibroblast-like synoviocytes,the high expression of CD44 mRNA in OA fibroblast-like synoviocytes may be not a result of inceased HA level of OA synovitis.
