Reactive oxygen species (reactive oxygen species, ROS) has been considered is a byproduct produced by the aerobic metabolism of plants , and may be anti - oxidant and antioxidant enzymes Clear . More recently, however, it was discovered that reactive oxygen species as signal molecules , play an important role in the process , such as plant growth , development, hormone signaling , response to biotic or abiotic environmental stress and programmed cell death . In this paper , a model compound of copper-zinc superoxide dismutase ( SOD mimetic enzyme ) soaked rice seeds under drought stress , the use of UV - visible spectroscopy , fluorescence spectroscopy , histochemistry , laser scanning confocal microscopy in vivo activity of rice the variation of the oxygen level . UV - visible spectroscopy analysis of the rice body's antioxidant enzyme activity levels change . First, using UV - visible spectroscopy , fluorescence spectroscopy rice plants H 2 sub > O 2 sub > O 2 < / sub > - < / sup · OH content of variation test , and found that the treatment groups were lower than the corresponding control group . Second, dimethyl benzidine (DAB) and nitro blue tetrazolium ( the nitroblue tetrazolium NBT ) histochemical staining method in rice leaves H 2 sub > O 2 < / sub > O 2 sub > - < / sup > levels in situ monitoring , found SOD simulate enzyme treatment to reduce the rice leaves under drought stress H < / sub> O 2 O 2 - sup> accumulation . Third, the use of a fluorescent dye H 2 sub > DCFDA and fluorescence microscopy and confocal laser microscopy rice root H 2 sub > O 2 sub > changes in the level imaging analysis found that SOD analog enzyme - treated rice roots H 2 O 2 content , lower than the corresponding control group H 2 O 2 < / sub> content . Fourth, the use of UV - visible spectroscopy rice leaf extract antioxidant enzyme activity in the liquid is determined . The study found that under drought stress conditions in the treatment group four antioxidant enzymes : SOD , CAT ( catalase ) , APX ( ascorbate peroxidase ) and GR ( glutathione reductase ) activity are high in the control group .
