Experimental Study of Radiotherapy on Echinococcus Multilocularis in Rats

Objective:To explore the effect of different 6-MeV ray on the Echinococcus multilocularis in rats. Methods:Setting up the model of secondary Echinococcus multilocularis infection in female SD rats, The rats were randomly divided into the following four groups, low dose radiotherapy group, moderate dose radiotherapy group, high dose radiotherapy group and control group, There were fifteen rats in each group. The rats were not accepted any treatment in control group.The rats were irradiated by 6-MeV ray in radiotherapy group, the total dose was 20Gy,40Gy and 60Gy respectively, and the single dose was 250cGy,500cGy and 750cGy respectively.There were eight times and the interval of radiation were three days, one month after the end of radiotherapy, The major judging indexes included wet weight of cyst, inhibition rate, histopathological changes of the cysts by both light microscopy and electron-microscopy. Results:The mean wet weight of cyst were (1.82±0.74) g, (1.04±0.38) g and (0.76±0.33) g, and the inhibition rate were 50%,72% and 82% respectively in radiotherapy groups. The mean wet weight of cyst was (3.65±0.79) g in control group, There were significant differences of mean cyst weights in the four groups (P<0.05). The light microscopy showed that the cysts of radiotherapy groups were different extents of alterations or damages, the structure was disorder, the corneous layer and germinal layer were degenerative or separated, the brood capsule and protoscolex was rare. The structure of cysts was normal in control group, the corneous layer and germinal layer showed clearly, There were protoscolexs in the brood capsule. There were significant differences of histopathological damages between radiotherapy groups and control group (P<0.05). The electron microscopy showed that the corneous layer and germinal layer were different extents of damages, and the damages of cysts were found most severely in the high dose radiotherapy group. Conclusions:Radiotherapy has effect in damaging the echinococcus multilocularis cysts and inhibiting multiplication of the protoscolex, and the effect is related to dose.
