Methological Study on Behavioral Observation in Pathological Model of Meridian Blockage in Minipigs

[Object] To probe into methods of observing behavioral indexes of experimental minipigs in the basic research on traditional Chinese medicine (TCM).[Medthods] In large animal house, the special cages and electronic surveillance system were adopted in this series to observe and record the changes in the behavior of minipigs before and after pathological modeling.[Results] The two set of experiments were conducted, the first group is divided into three experiments, the first one we choose 2 minipigs which appeare gastric and bowel bloating after their stomach meridian blocked by gel. In the second experiment, one pig which blocked gel in the stomach meridian appeares syndromes of stomach meridians, ingcluding not wanting to eat, appetite decrease, time for diet prolonged, defecate reduce, lassitude, mental status not good and so on, the other which blocked NS appeares no significant change. In the third experiment, we observed many quantitative indexes of 2 minipigs (one blocked gel and the other blocked NS) about their behaviour with N-of-1 statistics method, we found that 5 data--urine volume、stool weight、nocturnal number、nocturnal time、night foraging number appeared statistically significant after the experiment (no statistically significant before the experiment); And 6 data--night foraging time、daytime reposes number、daytime reposes time、daytime foraging number、daytime foraging time、average meal time appeared no statistically significant before and after the experiment; The number of climbing column appesred statistically significant before and after the experiment. In the second group, we observed 6 minipigs at the same time (No.1, 3,5 are blocked with gel and No.2,4,6 blocked with NS), N-of-1 statistics method shows that:Only No.3’s stool appesres statistically significant before and after the experiment. Urine and some other indexes show no statistically significant before and after the experiment. No.2,3,5,6 appear statistically significant before and after the experiment about feeding reaction grading. Only No.1’s meal time appesres statistically significant before and after the experiment. After mading pathological model for 17 days, No.2 and No.5 are chosen to give twice feed weight for 7 days, we found that their urine volume, feeding reaction、night foraging number have no statistically significant before giving the twice feed weight for 17 days, and come to statistically significant for 7 days after giving the twice feed weight, meal time appesres statistically significant before and after the experiment.[Conclusions] Research results show:Large animal house, new special cages and behavioral surveillance system can breeding and observation effectively for a long time, quantitative and grading records and calculation about many behavioral indexes laid a solid foundation in the methodology for Further Study about TCM animal diseases and pathological model from behavioral perspective.
