Purpose:Impatiens balsamina L. which is a well-known herbal medicine, recorded of Jiuhuang Bencao and Chinese Pharmacopoeia 1977, has been now widely cultivated as a medicinal and ornamental plant in China. The seeds of the plant have been used to treat lump in the abdomen and cancer of the esophagus, to act as an emmenagogue, to suppress puerperal pain, etc. The seeds have also been used as bactericide, anticancer agent. However, there is no comprehensive research of quality control of this drug, and the research of the anticancer effect of the active fraction is still imperfection. So, it is valuable to do the research on the seeds of Impatiens balsamina. We study on chemical constituents, microscopic character, TLC, checking, extract content and quantitive determination. To screen the anti-tumor activities of three extracts from the seeds of Impatiens balsamina through the method of pharmacodynamics in vitro; To optimize the excellent extraction technology of polysaccharides by response surface methodology and to screen the best method on deproteinization; To analyze the contents of the nine kinds of trace elements that are essential to human in the seeds by SPSS , and the results is going to be used to interpret the drug effect. To provid a reference for the quality standards, further development and utilization of the seeds of Impatiens balsamina.Materials and method:1. Screening experiment of anti-tumor activey of the extracts from the petroleum ether, ethyl acetate, n-butyl alcohol and polysaccharides.2.Through many kinds of methods such as silica gel, Sephadex LH-20, recrystallization, thin layer chromatography. Structure identification by physical and chemical identification spectroscopy, reference documents.3. By the method of herbalogical textual research, the original plant identification, Morphological, microscopic indentification, thin layer chromatography(TLC), checking and Extract determination. Determination of Quercetin and Kaemferol in the seeds of Impatiens balsamina from different area by the method of HPLC.4. The content of trace of elements were determined by ICP-MS, the correlation between the content of trace elements in Semen Impatientis were analyzed by SPSS and principal components analysis(PCA) was used to evaluate the quality.5. The independent variables were extraction temperature, extraction time and liquid-to-solid ratio, the dependent variable was extraction rate of polysaccharides in the seeds of Impatiens balsamina. Linear or nonlinear mathematic models were used to estimate the relationship between independent and dependent variables. Central composite design and response surface methodology were used to optimize the process of extraction. The prediction was carried out through comparing the observed and predicted values. Three methods include trichloroacetic acid (TCA), enzyme and enzyme-TCA were evaluated by the ratio of protein removing and polysaccharide removing. Box-Behnken Design-Response surface methodology was used to optimize the process of deproteinization.Results:1. The extracts from the ethyl acetate, n-butyl alcohol are active parts; Polysaccharides have the most remarkable effect of anticancer.2. They areβ-sitosterol (1),α-spinasterol(2), palmitic acd(3), Lawsone methyl ether (4),Quercetin(5),Kaempferol(6),Daucosterol(7),Stigmasterol(8).Among them, monomers palmitic acd(3), Lawsone methyl ether (4) and Stigmasterol(8)are obtained from the seeds of Impatiens balsamina for the first time.3. By the method of herbalogical textual research,the original plant identification, morphological, microscopic indentification, thin layer chromatography(TLC), checking and Extract determination. Determination of Quercetin and Kaemferol in the seeds of Impatiens balsamina from dif ferent area by the method of HPLC. systematic identification of pharmacognosy,providing the scientific evidence of identification and application.4. There was significant correlation between different elements. These elements take part in the pharmacodynamic functions of Semen Impatientis and the content of trace elements in different regions were significant differences .The results provide scientific basis for quality evaluation of the seeds of Impatiens balsamina.5. The optimum technological parameters were as follows: extracting temperature 92.0℃, extracting time 120.5min, solid -to- liquid ratio 1:25.8 and 2 times for extraction. The method of enzyme-TCA is the best mothod. The extraction technology is simple and reliable .
