Study on Relevant Cytokines in the Serum of Guinea Pigs Sensitized by Trichlorethylene

OBJECTIVE To investigate the skin sensitization effect of trichloroethylene on guinea pig and its mechanism, detect cytokines including TNF-α, IL-1β, IL-6, IL-8, IL-10 and IL-12 in serum of guinea pig, investigate the role of cytokines in TCE sensitized guinea pigs skin destruction, to explore the sensitization mechanism of TCE and skin destruction mechanism of Dermatitis Medicamentosa-like of Trichloroethylene(DMLT) by the research.METHODS Female guinea pigs, which were white and weighing 250～300g ,were adopted .After adaptive feeding a week, the Guinea Pigs were divided to Blank control group, solvent (olive oil) control group, TCE and DNCB group in random. To monitor the status of guinea-pig growth and health, their weight was measured regularly. Adopting Guinea Pig Maximization Test (GPMT) to establish sensitized guinea pig model, to observe and record the skin reaction in guinea pig back test area and take pictures. Reaction in guinea pig skin was scored in accordance with the skin reaction score standards in“Toxic chemicals identified technical specifications”(the Ministry of Health superintend made【2005】No.272). When the skin reaction score of test group≥1, it should be ruled positive skin sensitization, which was sensitized group, otherwise was non-sensitized group. 24h and 72h after sensitization test, suctioned the serum of guinea pigs and killed the animals. Serum were prepared for determining cytokine levels in serum , consisting of TNF-α, IL-1β, IL-6, IL-8, IL-10 and IL-12. Experimental results were analyzed with SPSS12.0. Results 1. In skin sensitization test, there were no erythema or edema in the blank group and olive oil control group; there were 15 animals showed erythema and edema reaction in DNCB positive control group,sensitization rate was 100%;18 of 29 animals in TCE test group appeared erythema, the average rate was 62.1%.2. TNF-αand IL-1βresults in guinea pigs serum: There were no significant difference between solvent control group and blank control group (P>0.05); and the level of TNF-αand IL-1βin TCE 24h sensitized group, TCE 72h group, DNCB 24h and DNCB 72h group were higher then it in solvent control group, and the difference was statistically significant (P <0.05); In TCE 24h and TCE 72h group, serum TNF-αand IL-1βlevels of the sensitized group were higher than those not sensitized, and the difference was statistically significant (P < 0.05).3. IL-6 and IL-8 results in guinea pig serum: Compared with control group, the IL-6 and IL-8 levels of solvent control group were no significant difference (P>0.05), and compared with solvent group, there were no statistically significant difference (P > 0.05) of serum IL - 6 and IL-8 levels in TCE 24h sensitized group, TCE 72h sensitized group, DNCB 24h and DNCB 72h group; The IL-6 levels of TCE 24h non-sensitized group were lower than 24h sensitized group, the IL-8 levels of TCE 72h non-sensitized group were lower than 72h sensitized group, and the difference was statistically significant (P <0.05).4、Results of IL-10: There were no significant difference of IL-10 level between solvent control group and blank control group (P > 0.05);The level of Serum IL-10 in TCE 72h sensitized group, TCE 72h no-sensitized group and the group DNCB 72h were higher than solvent control group, and the difference was statistically significant (P <0.05);There was no statistically significant difference of IL-10 levels between TCE 24h sensitized group and TCE 24h no-sensitized group, TCE 72h sensitized group and non-sensitized group (P > 0.05); 5、Results of IL-12: There were no significant difference of IL-12 level between solvent control group and blank control group (P > 0.05); Compared with solvent control group, the levels of serum IL-12 in each test group(TCE24h sensitized group, TCE24h no-sensitized group, TCE72h sensitized group, TCE72h no-sensitized group, DNCB24h and DNCB72h group) were lower, and the difference was significant(P > 0.05). Conclusion 1. TCE can sensitize guinea pigs, and sensitization rate was 64.29%.2. Cytokines TNF-α, IL-1β, IL-10 and IL-12 participate and play an important role in TCE skin allergic reaction.3. Cytokines IL-6 and IL-8 play a certain role in different period of TCE allergens reactions.
