Expression and Clinical Significance of Thrombospondin-1 and Transforming Growth Factor Alpha in Transitional Cell Carcinoma of the Bladder

Objective: To investigate tumor angiogenesis in bladder transitional cell carcinoma (TCCB) with thrombospondin -1 (TSP-1) and the significance of the expression of transforming growth factor- α (TGF-α) and its relationship with tumor microvessel density (MVD) . Method : Collect the Second Hospital of Lanzhou University, September 2007 to May 2009 menstrual open bladder tumors total resection or partial resection specimens and transurethral bladder tumor transurethral surgical specimens , postoperative pathology confirmed transitional cell carcinoma of paraffin-embedded specimens 85 cases , exclude adenocarcinoma , squamous cell papilloma . In another 32 cases of normal bladder tissue as control , application of immunohistochemistry to detect specimens TSP-1, TGF-α expression , while CD34 marker of microvascular tumor MVD count and analyze its relationship with clinicopathological parameters . Results: TSP-1 positive expression rate of bladder transitional cell carcinoma and normal bladder mucosa were 30.6% ( 26 / 85 ) and 65.6% ( 21 / 32) , a significant difference ( P lt; 0.01 ) , and TCCB pathological grade , clinical stage and tumor metastasis was negatively correlated with expression in bladder transitional cell carcinoma and normal bladder tissue ; TGF-α positive expression rate was 74.1% ( 63 / 85 ) and 21.9% (7 / 32 ) (P lt; 0.01), and pathological grading and clinical staging of bladder transitional cell carcinoma and tumor metastasis was no significant correlation ; MVD in bladder transitional cell carcinoma quantitative [ ( 66.54 ± 16.19 ) / mm2] , significantly higher than normal organization [ ( 43.56 ± 11.62 ) / mm2 ( P lt; 0.01 ) . Conclusion: TSP - 1 in bladder transitional cell carcinoma and normal bladder tissue compared to expression down and negatively correlated with pathological grade , clinical stage and tumor metastasis . TGF-α expression in bladder transitional cell carcinoma is enhanced . MVD values ??were higher than normal bladder tissue . Prompted the abnormal expression of TSP-1 and TGF-α are bladder transitional cell carcinoma angiogenesis inhibitor and accelerator , is closely related to factors involved in the regulation of tumor angiogenesis . Both jointly involved in the regulation of tumor development and expected to become the new target of the assessment of bladder transitional cell carcinoma of the degree of malignancy and treatment .
