Objective: To investigate further the understanding of the pathogenesis of depressive disorder of GSK-3β gene intronic polymorphism rs6438552 first episode major depressive disorder association between patients resting state fMRI. Methods: screening of major depressive disorder and normal controls in strict accordance with the inclusion criteria, gather general demographic information. 17 Hamilton (Hamilton Depression Scale, HAMD17) assessment of the severity of depression; application of polymerase chain reaction (PCR) and direct DNA sequencing technology, detection of GSK-3β single nucleotide polymorphism (SNP), according to GSK-3β the genotype grouping of; resting state scan all subjects; using professional data processing software to scan data preprocessing; the Reho value calculated for each patient using REST software and 3.0T MR scanner ALFF value, and final use spm5 software sample-to-sample t-test. Results: 1. General demographic data analysis age, gender differences between cases and controls had no statistical significance (P gt; 0.05) samples are comparable. 2. Hardy-Weinberg the genetic equilibrium test results of GSK-3βrs6438552 genotype distribution in each sample in Hardy-Weinberg genetic balance (P gt; 0.05), the selected sample group representation. 3 cases and control groups each genotype comparison between (1) the case and control groups ReHo compare: ① case group C / C genotype carriers with the control group, the C / C genotype was found in case group left forehead Back to ReHo value is higher than the control group, the the right suboccipital ReHo value than the control group to reduce; ② case group C / T genotype with the control group C / T genotype results not found to have special significance of brain regions; ③ case group T / T genotype carriers with the control group, the T / T genotype comparison the case group left inferior frontal gyrus, back to ReHo value higher than that of the control group and the right amount of the right temporal gyrus, left medial frontal gyrus ReHo low in the control group. (2) in case group and control group Alff analysis: ① C / C genotype of the patient group with the control group, C / C genotype Alff found that the case group right cingulate the Alff value compared to the control group increased ; (2) cases of group C / T genotype carriers and control group C / T genotype Alff comparison found that the patient group right temporal gyrus, right cingulate Alff value higher than that of the control group, the left middle frontal gyrus, left temporal gyrus, left postcentral gyrus and other brain regions the Alff value lower than that of the control group; ③ case group T / T genotype with the control group, T / T genotype Alff value comparison, the results found that the patient group left medial frontal gyrus the Alff value lower than the control group. Grouped according to the different genetic patterns results (1) ReHo results display case group C / C genotype had the right cerebellar anterior lobe ReHo value significantly higher than the C / T and T / T genotypes combined group, the control group, C / C genotype groups left cerebellar caudal lobe, left occipital and left frontal gyrus ReHo value compared with the C / TT / T gene merger group; (2) Alff fruit display cases of group C / TT / T genotype merger group left frontal gyrus the left brain occipital lobe Alff value was significantly higher than the C / C genotype groups, the occipital lobe the Alff value compared with the control group C / C genotype group left brain increased C / TT / T merger group; (3) ReHo and Alff analysis of results The case group T / T carriers of the left inferior frontal gyrus, the left superior temporal gyrus and right frontal back the ReHo value and Alff straight higher than the C / C and C / T merger group. Conclusion: 1.T / T genotype there may be a wider range of abnormal brain function activities. 2.GSK-3βrs6438552 intronic polymorphism may be associated with resting state functional brain activity. 3.GSK-3β gene may play a role in the development of depressive disorder.
