Objective: In this study, dorsal root ganglion (Dorsal root ganglion, DRG) as a model, the application of different frequency electrical stimulation in the rat sciatic nerve distal to observe the electrical stimulation of GABA-mediated DRG neurons membrane potential depolarization role of electrical stimulation on the DRG nociceptive information transmission. : Freshly isolated rat DRG sciatic nerve specimens application intracellular microelectrode recording technique, given by the the Powerlab bio-signal acquisition and processing analysis system (stimulation pulse width from 0.02 to 5 ms stimulus intensity V) voltage stimulation, stimulus intensity higher than the threshold intensity. On this basis, to the pulse width 5 ms, the strength of 2 V, the frequency for 1 Hz, 2 Hz, 5 Hz, 10 Hz and 50 Hz pulse wave distal sciatic nerve in rats, observed stimulation before and after the GABA (3 x 10 -4 mol / L) DRG neurons mediated membrane potential to changes in the polarization response amplitude. Results: 1. Frequency of 1 Hz, 2 Hz, 5 Hz, 10 Hz and 50 Hz, respectively, stimulation after 2 min, 2 min, 24 min, 12 min and 2 min maximal inhibition, inhibition rates were 16.78% ± 3.64%, 19.80% ± 3.80 %, 24.68% ± 3.99%, 29.20% ± 3.36% and 20.44% ± 3.12% (n = 4 to 5); its inhibitory effect duration to stimulation after 24 min, 24 min, 48 min,> 72 min and 48 min, 10 Hz inhibition of the duration of the longest. 3. Conclusion: The frequency electrical stimulation of the sciatic nerve, can inhibit DRG neurons membrane GABA depolarization and 10 Hz electrical stimulation of the strongest inhibition. The mechanism may be electrical stimulation to activate the nociceptive Aδ fiber nociceptive information to upload. Electrical stimulation of the GABAA receptor phosphorylation and inhibition of GABA-mediated presynaptic inhibition, to promote excitatory neurotransmitter release, the easy introduction of nociceptive information, leading to pain.
