Objective:To investigate the expresion of Cyclooxygenase-2(COX-2)and BCL-2 in differentiated thyroid carcinoma (DTC), study the pathogenesis of COX-2 and BCL-2 in the DTC and their roles and significance in clinical prognosis.Methods:The expression of COX-2 and BCL-2 protein was determined with SP immunohistochemical analysis in 40 papillary thyroid carcinomas(PTC), 32 follicular carcinomas(FTC)and 18 thyroidal adenomas and conduct comparative analysis.Results:1.The expression of COX-2 and BCL-2 in different tissues The positive expression rate of COX-2 in PTC is eminently higher than that in adenomas(P<0.01), and the expression rate of COX-2 in PTC is eminently higher than that in FTC(P<0.01), and the expression rate of BCL-2 in PTC and FTC is eminently higher than that in adenomas(P<0.01), while there is no difference between the expression rate of COX-2 in FTC and adenomas.2.The relevance of expression of COX-2 and BCL-2 in PTC Among 24 COX-2 positive samples from PTC group, there are 24 samples with the expression of BCL-2; while among 2 BCL-2 negatvie samples, there are 2 samples without COX-2 expression, the Spearman relevant anlysis indicates that there is positive correlation between the expressions of COX-2 and BCL-2 in PTC (r=0.631). The expressions of COX-2 and BCL-2 in PTC has been shown to be positively correlated.3.The relations among COX-2, BCL-2 and the parameter of thyroid cancer clinical pathology The TNM stage of thyroid cancer is determined in accordance with head-neck tumor stage criteria established by UICC 6th Edition 2002. There is visible difference between the expression of COX-2 in PTC and FTC in terms of pathology (P<0.01). In the PTC lymph node metastasis negative group and low prognosis index group, the expression of COX-2 shows higher rate than in the comparative group(P<0.05). There is no relation between the expression of BCL-2 and pathologies, but, the expression rate in the metastasis group with lymph node is eminently lower than that in the metastasis group without lymph node(P<0.05).Conclusion:COX-2 and BCL-2 is highly related to the biological behavior and prognosis of DTC. Overexpression of COX-2 or BCL-2 play a very important role in FTC progression. Associated detection of COX-2 or BCL-2 may help to judge the malignant degree and prognosis of DTC.
