Objectives:To study the antiviral effects and mechanisms of glycosaminoglycan from oyster(O-GAG) in vitro and in mice infected with herpes simplex virus typeⅠ.Methods:1.The effects of O-GAG on vero cells infected with herpes simplex virus typeⅠ(HSV-Ⅰ),including the effect of inhibiting virus replication,direct killing and colligateion,were observed by the cytopathic effect(CPE) and the MTT colorimetry methods;and the toxic effect of O-GAG on vero cells is also observed.2.After establishing the mice models infected with HSV-Ⅰ,the effects of O-GAG on weight,life-span,mortality rate,the pathological changes of some impotrant organs were observed.3.The effect of O-GAG on the thymus gland and spleen index and the phagocytosis of peritoneal macrophages in mice infected with HSV-Ⅰwas measured by macrophages neutral red test,the effects of the O-GAG on splenic lymphocyte transformation of the proliferation by MTT methods was also observed,and the implication of the O-GAG on animal immune function was analyzed.Results:1.Compared with he control group,O-GAG(100μg / ml,50μg / ml,25μg / ml) could effectively protecte vero cells infected with HSV-Ⅰ,increased cell proliferate activity(P<0.01),inhibited virus replication and decreased cytopathic effect caused by HSV-Ⅰ,and it had also a dose-effect relationship.O-GAG in vitro had no direct killing on HSV-Ⅰ,and O-GAG(50-1600μg/ml) did not shown toxicity on vero cells.2.Treated with different doses of O-GAG(10mg,20mg,40mg/kg),the average weight of mice infected with HSV-Ⅰwas increased,the average survival time was significantly prolonged and the mortality rate was decreased.O-GAG(10rag,20mg,40mg/kg) also has a protective effect on organs of mice infected with HSV-Ⅰ. 3.Compared with the virus group,O-GAG(10mg,20mg,40mg/kg) could significantly increase the spleen index and thymus index(P<0.05,P<0.01);O-GAG could enhance the phagocytosis of mice macrophage and splenic lymphocyte trans - formation of the proliferation(P<0.05,P<0.01).Conclusions:O-GAG has certain anti-HSV-Ⅰvirus function in vitro and vivo.It’s mechanism may be related with the effect of promoting the immune function.
