Schisandra sphenanthera is a vine plant, belonging to genera Schisandra and family Schisandraceae, which is also an important Chinese herb medicine, with the function of preventing lung but keeping health, taking care of nephridium and heart, stopping diarrhea, and protecting Baogan. Schisandra sphenanthera contains two effective elements. This paper mainly in Schisandra vitro cell suspension culture propagation and the cultivation conditions of the system, to enhance central Schisandraceae cells in the culture of active ingredients yield, large-scale production of the theoretical basis.The report deals with the successful propagation of this species using in vitro techniques.The experiment with different explants of Schisandra established systematically its tissue culture and suspension systems. In addition, effective elements which in the callus and cell were determined by HPLC. The main results were as follows:1.The callus could be induced from the different explants of Schisandra.The appearance time of callus and the rate of callus induction was different with the different explants. The petioles and caudex were better than leaves for callus induction,the optimal medium is MS+2,4-D 0.5 mg/L+BA 0.2 mg/L.2.The optimal medium of callus differentiation is MS+CPPU 1.0 mg/L+IBA 0.2 mg/L+NAA 2.0mg/L, times of the callus differentiation could reach 5.10. But adventitious buds growing weaker, and root of poor quality and not suitable for use as root or transplanting seedlings, have to add a process to improve seedling rooting rate.The optimal medium for rooting was 1/4 MS+BA 1.0 mg/L+NAA 1.0 mg/L+AC 0.5 g/L, the rate of rooting can reach 93.48%, with an average 3.5 cm long roots. In the hardened culture, combination of vermiculite and perlite with a ratio of 1:1 was used for transplant substances,which of the survival rate is the highest.3.Schisandra suspended central cell growth curve in a "S" shape, the best harvest period of 24 d. The suspension culture conditions for growth:2 g/20 mL inoculation of fresh weight, rotation speed of 110 rpm, sucrose concentration of 30 mg/L, medium size is 20 mL/100 mL, the hormone combination for MS+2,4-D 0.5 mg/L+BA 0.2 mg/L, respectively. PH value in the training process before or after the drop, at the end of training, pH 5.3.4. Schisandra by HPLC Schisandrin in the leaves, callus and suspension culture cells Deoxyschizandrin and Schisandrin B and found two kinds of effective components in suspension culture cells in the highest, followed by callus leaves the lowest.5.The innovations in this study are as below:The first successful establishment of Schisandra the tissue culture system and suspension culture system, and to explore the Schisandraceae plants, callus and suspension cells in the active ingredient in the relationship.
