Objective: This study intends to discuss the platelet-rich plasma (platelet-rich plasma, PRP) on human periodontal ligament fibroblasts (human periodontal ligament fibroblasts, HPDLFs) proliferation and migration, as well as with human bone marrow stromal cells (human bone marrow stroma cells HBMSCs) co-cultured cell proliferation ability, to provide experimental data for the application of PRP in the treatment of periodontal tissue regeneration. Method: 1. Using tissue culture method were isolated and cultured in vitro PDLFs detect the expression of vimentin to determine the biological source; whole bone marrow culture isolated and cultured human bone marrow stromal cells; PRP provided by the Blood Center of Jiangsu Province. 2, the MTT assay was used to detect different concentrations of PRP (1%, 5%, 10%, 20%, 30%, 50%) on PDLF proliferative capacity; Transwell system determination of different concentrations of PRP (1%, 5%, 10 %, 20%, 30%, 50%) the HPDLFs transition effect, serum-free culture medium as a negative control, 10% fetal calf serum as a positive control. 3 HPDLFs and HBMSCs 1:1 inoculated to transwell system, different concentrations of PRP (5%, 10%, 20%, 30%, 50%) were added to the transwell in serum-free medium as a negative control, 10% newborn calf serum as a positive control, separate the PDLFs cells were cultured alone as the negative control co-cultured and cultured for 7 days in the MTT assay. Results: (1) by tissue explant separation culture of HPDLFs express vimentin, are mesoderm source of cells; whole bone marrow culture method separation cultured human bone marrow stromal cells showed a fibroblast-like appearance, good proliferation capacity in vitro . 2. 3d, 5d and 7d, various concentrations of PRP group compared with the negative control group cells were significantly increased (P <0.01), a 20% concentration group (P <0.01); various concentrations of PRP group at 7 days The training period showed a significant time-dependent manner. Compared with the negative control, the different concentrations of the PRP can significantly promote cell migration (p <0.01), the PRP effects at a concentration of 10% of the group (91.80 ± 2.39) (P <0.01). 3.7 days, the cell volume of various concentrations of PRP group was significantly higher than the negative co-culture control group and negative cultured alone control group (P <0.01); coculture group stronger proliferative capacity than cultured alone proliferation group (P <0.01) ; to the best 10% of PRP concentration group (P <0.01). Conclusion: PRP can promote human periodontal ligament fibroblast migration and proliferation of bone marrow stromal cells under culture conditions, PRP promoting the best of human periodontal ligament fibroblasts proliferation.
