Detection of HER-2 Gene Amplification by TMA and FISH in Breast Cancer

Objective: Fluorescence in situ hybridization (FISH) technology single for the detection of breast cancer HER-2 gene unrecognizable positive cells from invasive carcinoma or carcinoma in situ of the part , in order to more accurately detect breast cancer organization HER-2 gene amplification and expression , the project seeks to tissue microarray (TMA) technology with the FISH technique combined joint detection , to provide a reference for correct clinical breast cancer diagnosis and individualized treatment . Method: 1. Collected 30 cases have been diagnosed with invasive ductal carcinoma of the breast ( with or without carcinoma in situ ingredients ) wax block specimens . Collected specimens FISH and immunohistochemistry (IHC) staining , detection of HER-2 gene amplification and protein expression , and compare the correlation between the two . 3 in all cases made ??of tissue microarray and FISH detection , assessment of HER-2 gene amplification , and comparison ordinary slice of FISH results . Results : 1.30 cases of ordinary slice FISH detection of HER-2 gene amplification of eight cases , no amplification of the 22 cases , IHC detection of HER-2 protein positive expression (3) three cases of suspected positive ( 2 ) 20 cases , negative ( 1 and 0) 7 cases . IHC positive gene amplification rate of 66.7% ( 2 / 3 ) , suspicious positive gene amplification rate of 30% ( 6 / 20 ) , negative had no gene amplification IHC and FISH results in line with the rate of 73.3% ( 22 / 30 ) , both significantly correlated (p lt; 0.01). By tissue microarray FISH detection , including detection success and the complete results in 13 cases , including the amplified 5 cases , no amplification of the eight cases , in line with the FISH results of ordinary slices was 76.9% ( 10 / 13 ) , two those related (p lt; 0.05), and the remaining 17 cases the organization off-chip , the organization not satisfied with the results . Our joint application Organization chip technology with FISH to detect breast cancer HER- 2 gene amplification , found that the method can be completely ruled out the FISH -positive cells in the carcinoma in situ HER- invasive breast cancer , which can be clearly identified 2 gene amplification . Conclusion: FISH and IHC detection of breast cancer HER-2 status results have relevance ; tissue microarray and FISH results of ordinary slices correlation ; joint application of tissue microarray technology and FISH technique can successfully and correctly detected the HER-2 gene amplification , and thus provide a more effective reference value for the correct clinical breast cancer diagnosis and individualized treatment .
