Effects of Qinxiang Granules on IFN-γ、IL-2 of Suckling Mice Infected with Human Rotavirus

Objective: To observe the Qin Hong particles on HRV infection of neonatal rat serum IFN-γ, IL-2 impact, to study the possible mechanism of of Qin Hong particle therapy HRV infection diarrhea, scientific theory and experimental basis for clinical application. Method: rotavirus oral infection model. 70 suckling mice stratified according to body weight and then randomly divided into seven groups of 10 each, one for the normal group, the other six groups caused by HRV infection model, model group, high dose group of Qinxiang Qinxiang middle dose group and low dose group Qinxiang ribavirin group Changkang piece group of seven groups. Regular feeding of the normal control group, and the remaining six groups were fed with normal saline, Qin Hong particles, low dose, ribavirin and intestinal Tablets, 3 times a day for consecutive 5d. Observe the general situation of suckling mice; the decapitated blood detected by ELISA in serum IFN-γ, IL-2 levels; colloidal gold assay feces of HRV antigen; HE stained light microscope to observe the pathological changes of the small intestinal cells. Results: ① modeling 12h colloidal gold assay detection of neonatal rat feces HRV antigen, model group, negative to positive, normal control group; ② general observation: HRV infection model group suckling mice reduced activity, diarrhea, weight gain slow or decline; normal group without diarrhea, rapid weight gain. Suckling mice, compared with the model group, each dose group generally better, including the high-dose group Qinxiang diarrhea least. ③ The suckling mouse droppings HRV antigen seroconversion rate: model group negative rate of 0, ribavirin group was 87.5%, Qin the incense high-dose group 75%, Qin Hong dose group 62.25%, 37.5% of the low dose group Qinxiang , 62.5% intestinal Kang slice group, the groups showed no significant difference (P gt; 0.05). ④ serum INF-γ content: Compared with the normal group, the parties modules have significantly increased, the difference was significant (P lt; 0.01); the piece group compared with the model group, Changkang, Qinxiang middle dose group, Qin the incense high-dose group can significantly increase the serum IFN-γ content (P lt; 0.05), Qinxiang low dose group differences is not obvious (P gt; 0.05). Qin Hong, the high-dose group significantly improve the ability of serum IFN-Y better than Changkang piece and ribavirin group (P lt; 0.05). ⑤ serum IL-2 levels: compared with the normal group, the model group were significantly higher (P lt; 0.01); compared with the model group, the Changkang piece group, ribavirin group, Qinxiang high, dose groups significantly (P lt; 0.05), and Qin the incense high-dose group was better than ribavirin Changkang piece group (P lt; 0.01). (6) cell morphology observation showed no abnormal: normal group, model group, small intestinal villi damage, inflammatory cell infiltration, intestinal epithelial cells in varying degrees in each dose group lesions, but lighter than the model group. Comparison of drug-treated group, the high dose group of Qinxiang reduce the role of the degree of intestinal epithelial cell vacuoles superior to the ribavirin group Changkang piece group. Conclusion: ① HRV infection by mouth, can cause to HRV infection diarrhea model; the ② Qinxiang particles of different dosages could improve HRV infection of suckling mice generally alleviate diarrhea symptoms, high doses Qinxiang role better than ribavirin and intestinal Tablet. The ③ particles may Qinxiang inhibit HRV infection of virus antigen in the neonatal rat feces. ④ Qin Hong particles can be enhanced the HRV infection neonatal rat serum IFN-γ, IL-2 expression, and render a certain dose-effect relationship, which may be part of the way to Qin Hong particle treatment HRV infection diarrhea. ⑤ Qin Hong particles can reduce small intestinal damage and inflammatory cell infiltration, may be one of the mechanisms for its treatment of HRV diarrhea
