Effect of Curcumin on Acute Lung Injury in Severe Acute Pancreatitis in Rats

Objective: To investigate the severe acute pancreatitis (severe acute pancreatitis, SAP) the role of the acute lung injury (acute lung injury, ALI) and its possible mechanism of curcumin (Curcumin). Methods: 45 healthy and clean SD rats were randomly divided into group A (sham group) 15 15 15 C B group (SAP) group (SAP curcumin intervention group). Retrograde cholangiopancreatography puncture injection to improve with 5% sodium taurocholate induced SAP and related ALI model. The three groups of rats at 6 hours after Caesarean section again. The naked eye abdominal cavity and pancreas general pathological changes. Blood test arterial partial pressure of oxygen (PaO2), partial pressure of carbon dioxide (PaCO2), and serum amylase activity. Collection of pancreatic tissue slices observed. Extend the incision into the chest, the naked eye broad chest and lung pathological changes. Take the left lung, estimated lung wet / dry weight ratios. Take the right lung, paraffin specimens slice observation: I hematoxylin - eosin (HE) staining, microscopy to observe and score; immunohistochemical SABC method detects nuclear factor-κB (nuclear factor kappa binding of NF-κB) protein expression and semi-quantitative analysis. Results: After 6 hours B, C rats pancreas gross microscopy was SAP change and lesions of group C than in group B alleviate. 6 hours after B, C rats lung gross, microscopic observation was ALI change, and the lesions of group C than in group B mitigate. 3 three groups of rats 6 hours after endoscopic lung tissue pathology score were: group A: 0.33 ± 0.49, B group: 7.33 ± 0.98, C: 5.27 ± 0.80; Group A the lt; B group, C group (P lt; 0.01), group B gt; C group (P lt; 0.01). 4 three groups of rats after 6-hour serum amylase activity were: Group A: 1063.20 ± 112.39 U / L, group B: 7319.27 ± 201.37 U / L, Group C: 4252.87 ± 199.41 U / L; including Group A lt; B group, C group (P lt; 0.01), group B gt; C group (P lt; 0.01). 5 three groups of rats after 6 hours PaO2 were: group A: 95.67 ± 2.97 mmHg, group B: 54.47 ± 3.34 mmHg, group C: 73.47 ± 4.03 mmHg; Group A gt; B group, C group (P lt; 0.01), B group lt; C group (P lt; 0.01). 6 three groups of rats after 6 hours PaCO2 were: Group A: 37.40 ± 2.90 mmHg, and group B: 54.73 ± 3.24 mmHg, group C: 45.00 ± 2.48 mmHg; Group A lt; B group, C group (P lt; 0.01), group B gt; C group (P lt; 0.01) the 7 three groups of rats 6 hours after the lung wet / dry weight ratios were: A group: 4.13 ± 0.34, B group: 9.05 ± 0.39, Group C: 6.14 ± 0.28 mmHg; Group A lt; B group, C group (P lt; 0.01), group B gt; C group (P lt; 0.01). The positive expression rate of NF-κB p65 in the lung tissue of 8 three groups of rats after 6 hours, respectively: group A: 0%, group B: 100%, group C: 60%; Group A lt; B group (χ to 2 = 5.96 P lt; 0.01) in group A the lt; C group (χ ~~ 2 = 4.92 P lt; 0.01), group B gt; C group (χ ~~ 2 = 2.07 P lt; 0.05). 9 6 hours after B, C lung tissue showed a significant positive correlation between expression of NF-κB and the severity of lung injury (B group: rs = 0.589 P lt; 0.01; C Group: rs = 0.803 P lt ; 0.01). Conclusion: 1 SAP rats ALI, lung tissue, there is overexpression of NF-κB, positive correlation between the severity of lung disease, suggesting that NF-κB APALI development process plays an important role in . 2 has a protective effect of curcumin on rats with SAP ALI mechanism of inhibition of NF-κB expression in lung tissue.
