Objective: To study different factors on hawthorn extraction rate and adsorption of macroporous resin purification and separation process, the application of single factor analysis with orthogonal experiment method of combining the optimization of the extraction and purification process to obtain the optimum extraction, purification and separation process route and acute liver injury in mice induced by alcohol, anti-inflammatory effect of hawthorn flavonoids. The use of single-factor analysis method: 1. Examine the solid-liquid ratio, extraction time, ultrasonic power, ethanol concentration of hawthorn flavonoids extraction process, and orthogonal experiment to determine the optimal conditions. 2 using single factor analysis examines flow rate, temperature, ethanol concentration, pH, hawthorn flavonoids purification process, and to determine the optimal conditions by orthogonal test. Established murine alcoholic liver injury model, of hawthorn flavonoids studied animal models of alcoholic liver injury in mice serum ALT, AST and liver tissue SOD, GSH-PX, NO, MDA, TNF-α. Results: 1 through single factor experiment to determine the impact of various factors conditions by orthogonal experiment to determine the optimum ultrasonic extraction conditions: A3B2C3D3: 85% ethanol as solvent, plus 15 times the solvent power of 700W ultrasonic assisted extraction 40min. By single factor experiment, the D101 is the best purification resin, preliminary experiments to select the temperature, pH, flow rate, eluent orthogonal test results to determine the best purification process conditions: A3B3C3D2, namely: 75% ethanol as the eluent, 80 ℃, pH 7.5 under the conditions of a flow rate of 2.0mL/min elution was carried out. Alcohol-induced animal models of alcoholic liver injury, hawthorn flavonoids through different doses of treatment, the results show that different dose the hawthorn can significantly reduce serum ALT, AST content (P lt; 0.05), increased liver tissue GSH-PX, SOD content (P lt; 0.05), to reduce the level of MDA, NO, TNF-α (P lt; 0.05), hawthorn flavonoids on alcoholic liver injury has a protective effect. Conclusion: Compared with traditional methods, the average yield of 4.736 percent, better extraction efficiency, ultrasound-assisted extraction of the Act can be significant savings extraction time, lower extraction costs, and have a good extraction rate. Macroporous adsorption resins enrichment and purification, the purity of 81.37% and 79.12% recovery rate, the method can be purified hawthorn flavonoids. Hawthorn flavonoids have a protective effect of alcohol-induced animal models of alcoholic liver injury may be related to reduce serum levels of free radicals, inhibition of lipid peroxidation.
