Ubiquitin (ubiquitin) is widely present in eukaryotes, contains 76 highly conserved amino acid residues. The ubiquitin gene ubiquitin fusion gene polyubiquitin gene in two forms. Ubiquitin gene and protein post-translational modifications, protein degradation, gene transcription, signal transduction, and secondary metabolite formation in fungi. Monascus (Monascus) is a typical filamentous fungi, there is documented as early as a thousand years ago, the Chinese classics. Monascus can produce a variety of secondary metabolites with important physiological activity. However, some studies have found that most of the red yeast strains in the fermentation process will produce a fungal toxins harmful to humans and animals - citrinin. If analysis clearly related to the the Monascus production citrinin gene using genetic engineering to build the citrinin strains that do not produce, you can fundamentally solve the problem citrinin the Monascus production process. The ubiquitin genes may be related to with Monascus production citrinin. Therefore, this thesis orange Monascus AS3.4384 is ubiquitin gene and its flanking sequences, specific research content and results are as follows: 1 by Southern hybridization with orange Monascus AS3.4384 cDNA subtractive Library Y- 130 ubiquitin gene EST fragment as a probe, and of Monascus orange AS3.4384 genome DNA restriction fragment into Southern hybridization, tapping recycling target fragment cloning, sequencing and analysis. The research results show that the orange red the aspergillosis AS3.4384 contain polyubiquitin gene and the ubiquitin fusion gene. Analysis the Orange Monascus AS3.4384 ubiquitin gene flanking sequences. Analysis of the experiments containing ubiquitin gene U2, U3, U6 three clones, the results show that the U2 in the ubiquitin gene downstream of a cell spindle during mitosis and chromosome centromere Blast x program combined with related checkpoint protein kinase gene; U3 ubiquitin gene downstream cell polarity of growth-related cell polarity protein gene; the ubiquitin genes downstream U6 with DNA replication and repair of sister chromatid binding protein gene. 3 Sau3AI digestion of Monascus aurantiaaeus AS3.4384 genome, using recycled 2-4 Kb fragments using pUC119 as a vector construct the orange red aspergillus AS3.4384 of genomic library, over 25,000 clones screened containing ubiquitin gene 3 positive clones, provides a technology platform for the analysis the Orange Monascus AS3.4384 ubiquitin gene and other genes.
