Objective: On the base of former pharmic effect experiment and clinical study, which have proved that Zhongjiling tablet can cure MG effectively, we will discuss the pertinence between pathological mechanism of Chinese medicine of “Qiyang is impotent, Zhenyuan is decadent, and Luoqi is weak and stagnant”and abnormity of immune function. We also will clarify immunity regulative function of Zhongjiling tablet, whose therapeutic principle is “warming and coordinating Qi yang, supporting Zhenyuan, and circulating Luoqi”. A new means and approach for the study of Chinese traditional medicine dealing with antoimmune disease will be offered . Method: Academic exploring: This thesis discussed the immunological mechanism of Zhongjiling tablet resisting MG in Qijing theory and Luobing Lysenko. “Qiyang is impotent, Zhenyuan is decadent”is the root of MG happening and “Luoqi is weak and stagnant”is the sign. Through relativity between protective functions of “Yuanqi and Weiqi”and western medicinal immune function, this thesis discussed the relation between “Qiyang is impotent, Zhenyuan is decadent”and abnormity of immunological function, and pertinence between “Luoqi is weak and stagnant”and N2AchR reducing and AchRab increasing. Experimental data scientifically elucidated that Zhongjiling tablet had double-side immune regulative function. Experimental research: 1.Effect of Zhongjiling tablet on immune function of normal mice: We divided female C57 mice into 5 groups at random: blank comparison group, dexamethasone group, cyclophosphamide group, Zhongjiling high dose group, and Zhongjiling low dose group. All the mice were poured stomach with medicine except blank comparison group and cyclophosphamide group. Blank group was supplied with equality volume’s menstruum, and cyclophosphamide group was injected cyclophosphamide in abdomen every other day. We poured the mice for 10 days in series. Then all the mice’s thymuses’and spleens’weight indexes, proliferous levels of T and B lymphocytes mediated by ConA and LPS, and the content of IL-2 stimulated by ConA were determined. 2.Effect of Zhongjiling tablet on immune function of immunosuppressive mice: C57 mice were divided into blank comparison group, cyclophosphamide model group, Zhong jiling high dose group, and Zhongjiling low dose group. Model mice were injected cyclophosphamide in abdomen on the first day, and other mice were injected water. Experimental groups were poured stomach with medicine and blank group was given menstruum for 10 days. Observed number and methods were all referred to anterior experiment. 3.Study of Zhongjiling tablet on immune regulative function of EAMG rats: According to thereference, we duplicated the EAMG model. We divided female Lewis rats into 5 groups at random: CFA group, model group, dexamethasone group, Zhongjiling high dose group, and Zhongjiling low dose group. All animals were given medicine or menstruum in the day of establishing model. Except for CFA group, all the rats were immunized with puried N2AchR for three times in general. We determined proliferous levels of T lymphocytes mediated by ConA and N2AchR.Then ELISA and RT-PCR were adopted to determine IFN-γ, IL-4, TGF-βlevels and mRNA expression. 4.Effect of Zhongjiling tablet on cellular superficial molecules of EAMG mice: C57 mice were divided into blank group, model group, Zhongjiling high dose group, and Zhongjiling low dose group. Except for CFA group, all the mice were immunized with puried N2AchR on the first day and the seventh day. These animals were given medicine or menstruum in the day of establishing model. After 10 days, cellular superficial molecules of mice were determined. Results: 1.Effect of Zhongjiling tablet on immune function of normal mice: Compared with blank comparison group, dexamethasone and cyclophosphamide were able to observably reduce mice’s thymuses’and spleens’weight indexes, P<0.01;Zhongjiling groups had no remarkable difference, P>0.05.
