Molecular Evolution and Phylogenetic Analysis of Some Species of Catantopidae (Orthoptera: Caelifera: Acriodidea) in China Based on Partial Sequence of Mitochondrial COⅡ Gene

Locusts and pastoral forestry pests, some species may cause serious harm to the deepening of locusts basic theoretical research, including the locusts system studies is essential. In recent years, the research into the level of molecular systematics. The Catantopidae is a Division of Acridoidea currently the molecular phylogeny Catantopidae not deep enough, and the need to use more molecular markers to explore the phylogenetic relationships of the Section. Mitochondrial coding gene cytochrome c oxidase subunit Ⅱ (CO Ⅱ) has become a school in the most widely used molecular markers of animal molecular systems, especially in insects. In this study, using PCR direct sequencing method for the determination of Catantopidae 10 subfamilies, 16 genera and 22 species of CO Ⅱ gene 585bp fragment, short nose and locust leprosy Acrididae Sunan as outgroup use ClustalX1.83 for sequence alignment , MEGA V2.1 sequence composition analysis, assessment of phylogenetic signal and data sets, using NJ method in PAUP * 4.0b10 (PPC) MP Act, the ML law as well as in the Mrbayes V3.0 Bayeux Adams phylogenetic inference (BI) phylogenetic tree. Eventually get the following results: of CO Ⅱ of 1.22 kinds Catantopidae insect gene sequence fragments to form a strong performance AT content bias (69.2%), especially in the third codon up to 88.8%. Conversion and transversion ratio (TS / TV) and genetic distance dependent. The amino acid sequence consisting of 19 kinds of amino acids composition, the lack of Cys at, Ile, Leu, Ser, Thr, Asp used more frequently. 2 CO Ⅱ gene the 585bp sequence fragments constituted full data set and loci divided codon depending on the codon of the first, second and third position data set phylogenetic signal assessment, including replacement saturation analysis, tree length distribution analysis and PTP test, the results show that different sets of data have a certain phylogenetic information. Using NJ, MP, ML and BI were constructed of 22 species. Catantopidae phylogenetic relationship tree, there are some differences found that four methods of phylogenetic tree, this description of the data set in 22 kinds limited information available on the phylogenetic relationships of the locusts.
