Cloth and β- elemene combined administration of anti-tumor effect and mechanism of celecoxib

ObjectiveMeasure the effect of Celecoxib &β-Elemene on proliferation inhibition and apoptosis induction of human hepatocellular carcinoma cell line HepG2, and reserch its possible mechanism. Detarmine the effect of combination therapy, provide a theoretical basis for further study.MethodsThe carcinoma cells HepG2 were treated with Celecoxib and elemene singly and in combination. Cell proliferation was determined using MTT assay; Apoptosis was detected using AO/EB douby staining with fluorescence microscope;cell apoptosis and the cell cycle was determined using FCM; Expression of apoptosis-releted proteins, COX-2, p38and P-p38 was determined using Western Blot. ResultsClecoxib or elemene alone could significantly inhibited HepG2 cell droliferation in a dose-dependent manner, and the inhibitory effect of Celecoxib combined with elemene was a joint synergy, The results of FCM showed the cell cycel was blocked in G2 phase. AO/EB doubly staining showed the cells appered typical characteristics of apoptosis. FCM showed that both of the two drugs induced HepG2 cells apoptosis. and has a synergistic effect. Results of Western Blot showed that the expression of Bcl-2, COX-2 was decreased; Bax, Caspase-3 expression increased; in the case of p38 expression unchanged, p38 phosphorylation levels rised. ConclusionBoth of Celecoxib and elemene can inhibit cell proliferation and promote cell apoptosis of human hepatocellular carcinoma cells HepG2,and combined use shows a synergic effect.
