The Pathogenic Role of Macrophage Migration Inhibitory Factor in Rat Acute Pancreatitis

Background & Aims: Macrophage migration inhibitory factor (MIF), originally described as an inhibitor of the random migration of macrophage, has gotten more attention. Recently, roles of MIF have been reported in the pathogenesis of several inflammatory diseases such as sepsis . The aim of this study was to explore the role of MIF in the pathogenesis of acute pancreatits, observing MIF expression in acute pancreatitis rats. Methods: Acute edematous pancreatitis (AEP) and acute necrotizing pancreatitis (ANP) were induced by the subcutaneous injection of cerulean into nap and by the injection of sodium taurocholate via retrocatheteris of common bile duct, respectively , on male Wistar rats. Blood samples and ascitic fluids in acute pancreatitis rats were collected from the inferior vena cava and the abodominal cavity , respectively, at 0 , 1 , 2 , 4 , and 8 hours after the induction of pancreatitis. MIF levels of all samples were determined by enzyme-linked immunosorbent assay (ELISA). Serum amylase levels in blood samples were measured. Immunohistochemical analysis for MIF was performed on the frozen sections of lung tissues from normal rats and rats with ANP (at 4 hr after the induction). Results: (1) Compared with those of normal rats ,serum MIF levels in AEP rats were increased (p<0.05) ,and serum and ascitic MIF levels in ANP rats were also increased (p<0.01) . (2) Ascitic MIF levels in ANP rats were increased rapidly with peaking around 1hour and sustaining the high level thereafter. Compared with serum MIF levels of ANP rats, ascitic MIF levels in ANP rats significantly increased . (3) Serum MIF levels were higher in ANP rats than AEP rats significantly (p<0.001). (4) Serum amylase levels were higher in AEP and ANP rats than normal rats significantly (p<0.05).(5) MIF positive immunostaining in rat lung was observed predominantly within the bronchial epithial cells and endothielial cells of blood vessels ,but not within the alveolar cells, even in normal rats . The staining was increased in ANP rats (at 4 hr after the induction).Conclusion:(l)MIF expression in acute pancreatitis significantly increased, suggesting a role of MIF in the pathogenesis of acute pancreatitis. (2) MIF might be an early cytokine of the inflammatory cascade in the pathogenesis of acute pancreatitis. Ascitic MIF might contribute to the increased serum MIF levels. (3) MIF levels might be correlated with the severity of acute pancreatitis. (4) MIF might play an important proinflammatory role in the pathogenesis of panceatitis-associated lung injury.
