Effect of Periodical Pulsation on Signal Transduction about Microorganism of SSF

Signal transduction of microorganism would be effected , if there were environmental stimulation. In the paper, the environmental system was solid-state fermentation reactor with periodical pulsation, the aim production was cellulase, compared with static solid state, the effect of periodical pulsation on signal transduction of the microorganism have been studied by the method of molecular biology. The paper is aimed at providing some proves for the new designing principles for the solid-state fermentation reactor with periodical pulsation.Firstly, the effect of varying the extraction and purification conditions for the microorganism protein obtained from Solid State Fermentation and the effect of periodical pulsation on protein have been studied. The mechanism of solid-state fermentation with periodical pulsation is also discussed. The results show that when 3ml Tris-HCl buffer was added to the solid-state fermentation system containing ca. 1 g of microorganism ,the maximum amount of intracellular protein which could be extracted was about 3.8mg .The highest purify ,amount of native protein and clearest sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)pattern were obtained when the impure protein was precipitated with a three fold excess of acetone at -20°C for 2h . The recovery ratio of FPA activity is 83.6% , while the recovery ratio of CMCase activity is 67%. Compared with static solid state fermentation, periodic pulsation afford a highest protein mass; form ca.1g of fermentation microorganism , 5.3mg of intracellular protein (an increase of 34.6%) and 9.09mg of extracellular protein (an increase of 17.8%)were obtained on the sixth day of the fermentation . The FPA enzyme activity 、 CMCase enzyme activity and β-Glucosidase enzyme activity of extra cellular protein are 104.34 IU.g-1 、 6575.52 IU.g-1and 3.99IU.g-1 respectively, which represent increases of 60.1% 、 21.2% and 2.5% over the corresponding values for static solid state fermentation . The FPA enzyme activity and CMCase enzyme activity of intracellular protein are 14.7 IU.g-1 and 383.52 IU.g-1respectively, which represent decreases of 22.2% and 38.7% over the corresponding values for static solid state fermentation .The enzyme activity of the
