Study on the Effects of Mitomycin C Drug Delivery System in Experimental Glaucoma Filtration Surgery

Objective: To study mitomycin C delivery system (mitomycin C drug delivery system, MMC DDS) efficacy of the rabbit eyes filtration surgery after anti- scar formation and rabbit ocular tissue toxicity discussed as glaucoma filtration the feasibility and effectiveness of the anti- scarring after surgery adjuvant . Methods: (1 ) mitomycin C sustained release system : carrier material is polylactic acid ( , polylactic acid, PLA), and a polysaccharide acid (polyglycolic acid, PGA) , a copolymer of poly lactose acid ( polylactioglycolic - acid, PLGA ) , white a cylindrical shape, a diameter of 1.0mm, and a length of 2.0mm. Release agent mitomycin C by MMC (0.02mg) and carrier composed of dark gray , the same as the size and shape of the blank carrier , expected release time for four weeks . Ethylene oxide fumigation for 24 hours , place one week after the spare . ( 2) the establishment of experimental filtration surgery model : the random selection of 24 2.0-2.5kg healthy New Zealand white rabbits , of either sex , both eyes underwent trabeculectomy . MMC DDS 1 capsules implanted in all the the rabbits right eye surgery left eye randomly selected for the experimental group (MMC DDS group ) , 12 eye surgery implant blank drug delivery system ( empty drug delivery system , EM DDS ) Capsule as negative control (EM DDS group ) , 12 eyes for the conventional treatment group , positive control intraoperative 0.2mg/ml MMC . (3) were observed after the rabbit eye whether eyelid edema , conjunctival edema , anterior chamber inflammation and corneal toxicity to determine the tolerability of rabbit eyes MMC DDS . ( 4 ) postoperative intraocular pressure (IOP), bleb height and range , the concentration of the drug within the aqueous humor to determine MMC DDS effect of filtration surgery . (5) respectively after 7 days , 14 days , 28 days were sacrificed rabbit enucleated for histopathological examination of the surgical site , observed MMC DDS filtration after filtration surgery Road parts inflammatory response , fibroblast cell proliferation . ( 6 ) The application of immunohistochemical techniques to observe the surgical site fibroblast expression of proliferating cell nuclear antigen (PCNA) to explore the mechanism of scar formation after mitomycin C delivery system anti- glaucoma filtration surgery . Results: (1) rabbit eyes after filtration surgery MMC DDS showed good tolerability throughout the experiment
